Serine-rich single-pass membrane protein 1 (SSMEM1) is a conserved, testis-specific, single-pass transmembrane protein encoded by the human SSMEM1 gene (also known as C7orf45 and FLJ40316), located on chromosome 7q32.2[1]. The protein is 244 amino acids long and contains a domain of unknown function (DUF4636) spanning most of its sequence[1]. SSMEM1 is highly expressed in the testes and is upregulated during spermatogenesis, coinciding with the appearance of pachytene spermatocytes[2][3][4]. Functional studies in mice have shown SSMEM1 is essential for the proper formation of sperm head shape and sperm motility; knockout results in male infertility, globozoospermia (round-headed sperm), and loss of motility, due to abnormal intracellular Golgi migration during spermatid development[2][3][4][5]. SSMEM1 is currently not known to be targeted by drugs and is not considered a canonical therapeutic target, although its role in infertility could have future implications for contraceptive development.
