The serotonin 1A receptor (5-HT1A) is a subtype of serotonin receptors that binds the neurotransmitter serotonin (5-hydroxytryptamine, 5-HT). It is one of the most abundant and widely distributed serotonin receptors in the brain and plays a crucial role in modulating mood, anxiety, cognition, and various neuroendocrine functions. It functions as both an autoreceptor on serotonergic neurons and a heteroreceptor on other neurons, mediating diverse physiological effects. It is a major target for drugs treating mood disorders such as depression and anxiety.
Agonist binding inhibits adenylyl cyclase activity, decreases cAMP levels, opens inwardly rectifying potassium channels leading to hyperpolarization, and inhibits voltage-gated calcium channels to reduce neurotransmitter release. Autoreceptor activation provides negative feedback to limit serotonergic signaling.
