The 5-hydroxytryptamine receptor 1D (HTR1D) is a G protein-coupled receptor for serotonin that mediates inhibitory neurotransmission. It is primarily known for its role in migraine pathophysiology, where triptans act as agonists to induce vasoconstriction. It is expressed in brain regions, including the basal ganglia, hippocampus, and cortex, and in vascular smooth muscle cells.
Triptans act as agonists at both 5-HT₁B and 5-HT₁D receptors, leading to vasoconstriction of cerebral blood vessels, inhibition of vasoactive neuropeptide release, and suppression of nociceptive neurotransmission.
