The serotonin synthesis pathway is a metabolic route responsible for the biosynthesis of serotonin (5-hydroxytryptamine, 5-HT) from the essential amino acid L-tryptophan. This pathway consists of two main enzymatic steps: hydroxylation of L-tryptophan by tryptophan hydroxylase (TPH) to form 5-hydroxytryptophan (5-HTP), and decarboxylation of 5-HTP by aromatic amino acid decarboxylase (AAAD) to form serotonin. Serotonin produced via this pathway regulates diverse functions, including central nervous system neurotransmission affecting mood, sleep, appetite, and cognition, as well as gastrointestinal motility and secretion. Alterations or dysregulation within this pathway are implicated in various disorders. Tryptophan intake can influence brain levels of serotonin because tryptophan crosses the blood-brain barrier, while serotonin itself does not.
