Overview

The serotonin transporter (SERT) allosteric site is a regulatory region on the SERT protein, distinct from the primary substrate binding site. Located within the extracellular vestibule, it is primarily formed by transmembrane helices TM10–TM12 and extracellular loop 6 (EL6). Binding of ligands at this allosteric site modulates SERT function, influencing both substrate transport and inhibitor binding kinetics. Ligands such as serotonin and certain antidepressants can occupy this site, affecting the efficacy and duration of SSRIs and providing avenues for the development of novel antidepressants.
