Overview

Serpin family E member 2 (SERPINE2) mRNA encodes a secreted protein also known as Protease nexin 1, which belongs to the serpin superfamily of serine protease inhibitors (UniProt: P07093). The protein primarily targets and inhibits proteases such as thrombin, urokinase-type plasminogen activator (uPA), and plasmin, thereby regulating extracellular matrix (ECM) degradation and cell signaling (NCBI Gene: 5270). In oncology, SERPINE2 is often upregulated and functions as an oncogene by promoting tumor cell invasion, metastasis, and angiogenesis through the modulation of the tumor microenvironment (PubMed: 12644675). Targeting the SERPINE2 mRNA specifically allows for the knockdown of this protein, which has shown promise in preclinical models for reducing the metastatic potential of aggressive cancers like pancreatic and breast carcinoma. Additionally, genetic variants and expression levels of SERPINE2 are linked to the development of chronic obstructive pulmonary disease (COPD), where it influences lung function and airway remodeling (PubMed: 16775174). Therapeutic strategies focusing on SERPINE2 mRNA utilize antisense oligonucleotides or RNA interference to achieve highly specific gene silencing. While no clinical drugs are currently approved, the molecule remains a significant target for precision medicine in fibrotic and malignant diseases.
