Serpin peptidase inhibitor, clade E (nexin, plasminogen activator inhibitor type 1), member 3 (abbreviated SERPINE3) is a protein-coding gene that encodes a putative serine protease inhibitor belonging to the serpin superfamily[4][3][2]. SERPINE3 is expressed specifically in the retinal pigment epithelium (RPE) in humans and is secreted in extracellular vesicles[1]. It is predicted to function as a serine-type endopeptidase inhibitor and may be involved in regulating proteolysis within the eye, stabilizing neuronal connections, or protecting neurons from proteolytic damage in the retina[1][3][4][5]. Protease inhibition screening suggests potential targets include caspases 1, 2, and 5, as well as cathepsin K[1]. Knockout of Serpine3 in mouse models leads to structural retinal abnormalities, indicating a critical role in retinal integrity and RPE function[1]. No direct drug interactions, clinical biomarkers, or established therapeutic mechanisms are currently known.
