Serum-glucocorticoid kinase (SGK1) is a serine/threonine protein kinase belonging to the AGC family. It plays a central role in cellular stress response, regulating ion channels, transporters, cell survival pathways, hormone release, neuroexcitability, inflammation, cell migration, angiogenesis, wound healing, and metabolic processes. SGK1 expression is induced by serum, glucocorticoids, mineralocorticoids, gonadotropins, cytokines, and growth factors. Activation occurs via phosphorylation through PDK1 and mTORC2. Altered SGK1 expression/activity is linked to hypertension, diabetic neuropathy/nephropathy, ischemic injury, neurodegeneration, cardiac fibrosis/remodeling, and cancer progression/metastasis, making it a potential therapeutic target.
