Serum response factor (SRF) is a MADS-box family transcription factor that binds to the serum response element (SRE, also called CArG box) in the promoter region of hundreds of target genes[1][2][4][5]. It is essential for transducing signals from the cytoskeleton and extracellular environment to the nucleus, regulating numerous biological processes such as immediate early gene expression (e.g., c-fos), cell cycle progression, apoptosis, cell growth, muscle cell differentiation, development of the cardiovascular system, and cytoskeletal organization[1][2][3][4][6]. SRF interacts with key cofactors—most notably myocardin-related transcription factors (MRTFs) and ternary complex factors (TCFs)—and is the nexus for multiple signaling pathways, including Rho-actin and MAP kinase cascades[2][4][6]. SRF is implicated in several disease mechanisms, including cancer, cardiovascular disorders, and certain skin diseases like psoriasis[1][3][6]. Structural studies reveal a highly conserved DNA-binding and dimerization domain that integrates diverse regulatory inputs[5][7]. Although not a classical drug target, experimental modulation of SRF and its regulatory pathways remains an area of therapeutic research, with the potential for broad systemic effects[6].
