Overview

SET and MYND domain-containing protein 3 (SMYD3) mRNA is the messenger RNA transcript that encodes the SMYD3 protein, a lysine methyltransferase essential for epigenetic regulation and cellular signaling (UniProt Q9H7B4). The SMYD3 protein facilitates the methylation of histone H3 at lysine 4 (H3K4) and non-histone proteins like MAP3K2 and VEGFR1, thereby activating genes involved in cell cycle progression and oncogenic pathways (Hamamoto et al., 2004, Nature). As a therapeutic target, SMYD3 mRNA is the focus of nucleic acid-based strategies, including small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), designed to silence gene expression and reduce the abundance of the oncogenic protein (Liu et al., 2013, FEBS Letters). Overexpression of SMYD3 mRNA is a hallmark of several aggressive cancers, such as hepatocellular carcinoma, breast cancer, and colorectal cancer, where it serves as a biomarker for poor prognosis (Giakountis et al., 2017, Cancer Research). Targeting the mRNA transcript allows for the inhibition of SMYD3-mediated transcriptional programs that drive tumor growth and metastasis (Sarris et al., 2016, Gene). While small molecule inhibitors are being developed to target the protein's catalytic activity, mRNA-directed therapies provide an alternative approach to completely ablate SMYD3 function in malignant cells.
