Overview

SET domain-containing protein 9 (SETD9) is part of a large family of proteins characterized by the presence of a SET domain, which imparts lysine methyltransferase activity. These enzymes regulate chromatin structure and gene transcription by methylating lysine residues on histones, a process central to epigenetic control. The SET domain, named after three Drosophila proteins (Su(var)3-9, Enhancer of zeste, and Trithorax), forms a unique knot-like structure that is distinct from classic methyltransferase folds[1][4]. SET domain-containing proteins are implicated in essential processes such as cell proliferation, growth control, and differentiation. While specific roles of SETD9 in disease are still emerging, the SET domain family, in general, has strong links to cancer biology and other transcriptional dysregulation disorders[1][5]. SETD9 may become increasingly relevant as a drug target and biomarker in the future.
