The Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Omicron variant membrane (M) protein is a highly conserved structural protein that plays a central role in the viral life cycle by organizing the assembly of new virions. It is the most abundant protein in the viral envelope and acts as a scaffold, interacting with the spike (S), envelope (E), and nucleocapsid (N) proteins to drive viral budding and define the virus's pleomorphic shape. In the Omicron variant, the M protein possesses specific mutations, such as D3G, Q19E, and A63T, which distinguishes it from ancestral strains and may influence its ability to antagonize host interferon responses and facilitate immune evasion. Because it is highly conserved relative to the rapidly mutating spike protein, the M protein has emerged as an attractive therapeutic target for broad-spectrum antiviral agents. Research into small-molecule inhibitors, such as JNJ-9676 and CIM-834, has demonstrated that targeting the M protein can stabilize its dimeric structure in an inactive conformation, effectively blocking the assembly and release of infectious viral particles.
