The SARS-CoV-2 RNA genome is a ~29.9 kilobase, single-stranded, positive-sense RNA molecule that constitutes the entire genetic material of the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), the causative agent of COVID-19[1][2][3]. It is the largest known RNA genome among coronaviruses and encodes both structural proteins (including spike, membrane, envelope, and nucleocapsid) and non-structural proteins crucial for viral replication and pathogenesis[2]. The genome displays complex secondary and tertiary structural elements, including highly structured regions (stem-loops, pseudoknots, and long-range duplexes) that regulate translation, replication, and viral RNA synthesis[1][3][6]. These RNA structures are subject to evolutionary pressures and are essential for viral packaging, making specific regions of the genome promising targets for siRNA- and ASO-based antivirals as well as for the development of small molecules that perturb RNA conformation. The RNA can serve as a biomarker of infection, with its quantity in patient samples correlating with disease severity and stage. However, the high mutation rate inherent to RNA genomes and challenges in selective targeting and delivery complicate therapeutic intervention[2].
