Overview

SH2 domain–containing protein 3C (SH2D3C) is an adaptor protein found in the cytoplasm, characterized by an SH2 (Src homology 2) domain and a guanine nucleotide exchange factor–like domain[3]. It plays key roles in signal transduction processes involved in cell adhesion, migration, tissue organization, and immune response regulation. SH2D3C participates in integrin-mediated cell adhesion through the BCAR1-CRK-RAPGEF1 pathway, promotes cell migration and invasion through the extracellular matrix, and is critical for the development and function of B cells and T lymphocytes, especially those involved in the splenic marginal zone and peripheral tissues[3]. Multiple transcript variants and isoforms are described, with some isoforms specialized for chemokine-induced lymphocyte migration or participating in distinct signaling pathways. To date, SH2D3C is not recognized as a direct therapeutic target, and there are no known drugs or biomarkers directly associated with it[3].
