Overview

SH2 domain-containing proteins are a large family of intracellular signaling molecules characterized by the presence of one or more Src Homology 2 (SH2) domains. These domains are approximately 100 amino acids in length and play a critical role in cellular signal transduction, particularly within tyrosine kinase signaling pathways. SH2 domains specifically recognize and bind to phosphorylated tyrosine residues on target proteins. This binding mediates protein-protein interactions that transmit signals from activated cell surface receptors (often receptor tyrosine kinases) to downstream effectors, influencing diverse cellular processes such as gene expression, cell growth, differentiation, migration, and survival. Mutations affecting either the structural stability or binding properties of an SH2 domain can disrupt normal signal transduction. Such disruptions have been implicated in various diseases including immunodeficiencies and certain cancers. Because they mediate crucial protein-protein interactions in disease-relevant pathways—especially those involving oncogenic kinases—SH2 domains have become attractive targets for drug discovery.
