SH2 domain-containing protein 3A (SH2D3A) is a human protein encoded by the SH2D3A gene. It is an adapter protein featuring both an SH2 domain and a Ras guanine-nucleotide exchange factor (Ras-GEF) domain, enabling binding to phosphotyrosine residues and likely mediating interactions in signal transduction pathways, including possible involvement in the JNK cascade. SH2D3A interacts with proteins such as the epidermal growth factor receptor and belongs to a family of adapter proteins that couple phosphotyrosine signaling to small GTPase–mediated pathways. While SH2 domain proteins are broadly important in regulating cellular signaling, SH2D3A itself has no direct validation as a drug target or established roles in major diseases beyond weak associations with certain infectious encephalitides.
