SH2 domain-containing protein 6 (SH2D6) is a cytoplasmic adaptor protein characterized by its Src Homology 2 (SH2) domain[4][5]. This domain enables the protein to bind specifically to phosphorylated tyrosine residues on target peptides, thus participating in intracellular signaling cascades, particularly those downstream of cell surface receptor protein tyrosine kinases[1][3][4]. SH2 domain adaptor proteins are essential for mediating signal transduction, helping route information from receptors on the cell surface to downstream signaling elements within the cell[1][3][6]. SH2D6 shares similarity with other signaling adaptors, such as BLNK, but its precise physiological ligands, interaction partners, and biological functions remain incompletely characterized. Current evidence from gene databases links SH2D6 only to rare genetic disorders through gene association, with no established therapeutic target status and no interacting drugs or known safety, biomarker, or mechanism-of-action profiles[4][7].
