Overview

SH2B adapter protein 2 (SH2B2, also known as APS) is an adaptor protein highly expressed in B lymphocytes and notable for containing both pleckstrin homology (PH) and Src homology 2 (SH2) domains[1][5]. Upon B cell receptor stimulation, SH2B2 becomes tyrosine-phosphorylated, facilitating protein-protein interactions such as binding to Grb2, and Shc adaptor proteins, which are important for the integration and propagation of intracellular signaling.[1][5] SH2B2 is functionally implicated in multiple receptor tyrosine kinase signaling cascades, including responses to insulin, leptin, and cytokines, by acting as a molecular scaffold to connect activated kinases (such as TrkA and Cbl) with downstream effectors[1][5]. Its role in signaling makes it relevant to immune function and potentially oncogenesis, especially in hematopoietic cell lineages. There are no known selective drugs or inhibitors currently approved for clinical use that target SH2B2 directly.
