Overview

SH2B adapter protein 3 (SH2B3), also known as LNK, is an intracellular adaptor protein containing an SH2 domain, primarily expressed in hematopoietic and endothelial cells. It acts as a central negative regulator of cytokine and growth factor receptor signaling, especially pathways mediated by JAK2, JAK3, and the erythropoietin receptor. By acting as a molecular brake on cell proliferation, hematopoiesis, and immune responses, SH2B3 limits excessive signaling that can lead to blood cell expansion, inflammation, and autoimmunity. Variants and mutations in the SH2B3 gene are implicated in various conditions, including autoimmune diseases (such as type 1 diabetes and celiac disease), hematologic cancers, and cardiovascular diseases (notably coronary artery disease and hypertension). Although not a traditional receptor or enzyme, SH2B3 serves as a scaffold for protein–protein interactions, tuning signaling responses and representing a potential, though as yet unproven, therapeutic target in diseases of immune dysregulation, hematopoietic proliferation, and vascular inflammation
