Overview

SH3 and cysteine-rich domain-containing protein (STAC) is a member of the STAC adaptor protein family, present in isoforms such as STAC1, STAC2, and STAC3[2][4]. STAC proteins contain an SH3 domain and a cysteine-rich (C1) domain and play a key role in skeletal muscle physiology by mediating excitation–contraction (EC) coupling. STAC3 is crucial for the coupling of skeletal muscle L-type calcium channel (dihydropyridine receptor, DHPR) and ryanodine receptor 1 (RyR1), enabling the calcium release necessary for muscle contraction[1][2][4]. STAC protein dysfunction is associated with congenital myopathies and potentially predisposes to malignant hyperthermia[1][3]. STAC proteins act as auxiliary and regulatory proteins rather than traditional druggable therapeutic targets like receptors or enzymes, and to date, they are not known to be direct drug targets[2][3][4].
