Overview

SH3 and cysteine-rich domain-containing protein 2 (STAC2) is a protein coded by the STAC2 gene that contains both an SH3 domain and a zinc finger (cysteine-rich) domain[3][5]. STAC2 is part of the STAC family of proteins, which also includes STAC1 and STAC3, sharing a common domain architecture of an N-terminal C1 domain (a type of zinc finger) linked by a disordered region to tandem SH3 domains[2][3][4]. STAC2 is predominantly expressed in the central nervous system and acts as a regulator of L-type voltage-gated calcium channels, such as CACNA1C (CaV1.2)[2][3]. It slows voltage- and calcium-dependent inactivation of these channels, influencing neuronal excitability and calcium signaling[2][3]. While STAC3 plays a crucial role in skeletal muscle excitation–contraction coupling, STAC2 is primarily expressed in neural, reproductive, and endocrine tissues[2]. Disease associations for STAC2, while less direct than for STAC3, include possible contributions to heart failure via altered expression[3] and potential involvement in neurological processes via its modulation of calcium channels. No direct drug interactions or mechanisms of action have been described for STAC2, and its role as a direct therapeutic target is not currently established.
