Overview

SH3 and multiple ankyrin repeat domains protein 2 (SHANK2) is a postsynaptic scaffold protein and member of the Shank family, primarily localized at the postsynaptic density (PSD) of excitatory synapses in the brain[1][2]. SHANK2 contains ankyrin repeats, SH3, PDZ, proline-rich, and sterile alpha motif (SAM) domains, allowing it to organize large protein-protein interaction complexes and couple membrane receptors, such as NMDA-type and metabotropic glutamate receptors, to the cytoskeleton[1][2][3]. This structural framework is essential for the development, maturation, and function of dendritic spines and synaptic connectivity. Genetic variants in SHANK2 are strongly implicated in autism spectrum disorder, intellectual disability, schizophrenia, and related neurodevelopmental conditions[2][4][5]. While not a typical therapeutic target like a receptor or enzyme, SHANK2 is a key molecular hub in the PSD, and its dysfunction disrupts normal synaptic transmission and neural circuit organization.
