Overview

SH3 and PX domain-containing protein 2A (SH3PXD2A, also known as TKS5) is an intracellular adapter and scaffold protein containing five SH3 domains and a PX domain. It acts as an organizer protein required for the formation of dynamic actin-rich structures such as podosomes and invadopodia, which are involved in cell invasion and extracellular matrix degradation. SH3PXD2A is necessary for protease-driven invasion by cancer cells and plays a role in superoxide production via activation of NADPH oxidases. It also mediates some neurotoxic effects in association with amyloid-beta. This protein is highly expressed in invasive cancer cells and is involved in cell migration, invasion, and signaling pathways such as those regulated by Src-family kinases[1][2][4][6][7][8].
