Overview

SH3 and SYLF domain containing 1 (SH3YL1) is a cytoplasmic adaptor-like protein containing SH3 and SYLF domains, involved in phosphatidylinositol binding and phosphatase interaction. It localizes to the plasma membrane, regulates dorsal ruffle assembly, and participates in endocytosis and membrane remodeling. SH3YL1 has been identified as a regulator of NOX4, contributing to ROS production and implicated in kidney pathophysiology, including diabetic nephropathy and thrombophilia. It serves as a novel biomarker for renal outcomes in diabetic kidney disease, with higher plasma or urinary levels correlating to worse renal prognosis in type 2 diabetes. No direct therapeutics targeting SH3YL1 are known; its disease relevance is primarily as a prognostic/predictive biomarker rather than a therapeutic target.
