Overview

SH3 domain-binding protein 1 (SH3BP1) is an enzyme classified as a Rho GTPase-activating protein (RhoGAP), which regulates the activity of Rho-type GTPases, including RAC1 and CDC42, by accelerating the conversion of their active GTP-bound state to the inactive GDP-bound state. SH3BP1 modulates cytoskeletal dynamics, cell motility, actin organization, epithelial junction formation, and phagocytosis, often serving as a negative regulator in these processes. Its cellular functions support proper cell migration, morphogenesis, and responses to semaphorin signaling, with established roles in the exocyst complex and interaction with Plexin-D1 and Rac signaling. Overexpression or dysregulation of SH3BP1 can promote metastasis in human liver cancer, and it is genetically associated with Aarskog-Scott syndrome.
