SH3 domain-containing protein 21 (SH3D21) is a human nuclear protein encoded by the SH3D21 gene on chromosome 1p34.3[1]. It contains three Src homology 3 (SH3) domains, typically known for mediating protein–protein interactions, especially recognizing proline-rich motifs[1][3][4]. While its precise function is not fully characterized, SH3D21 is predicted to be involved in protein complex assembly and cellular signaling. It is expressed at low levels in most tissues but shows high expression in certain neuronal ganglia. SH3D21 has two protein isoforms differing in the number of SH3 domains. Biological studies suggest potential links to actin filament organization and cell migration (especially from model organisms), and changes in SH3D21 expression have been observed in specific disease contexts, though direct causative roles remain unproven. No interacting drugs, biomarker roles, or notable safety concerns have been reported to date for this protein[1][2][4].
