SH3BP5 antisense RNA 1 (SH3BP5-AS1) is a long non-coding RNA, transcribed antisense to the SH3BP5 gene on chromosome 3. It is upregulated in pancreatic cancer and associated with resistance to gemcitabine chemotherapy, poor prognosis, and increased malignancy. SH3BP5-AS1 acts as a competing endogenous RNA, sponging microRNAs (notably miR-139-5p), and upregulates CTBP1 to activate the Wnt signaling pathway. Its expression is regulated through N6-methyladenosine (m6A) RNA modifications. SH3BP5-AS1 is being explored as both a biomarker and a potential therapeutic target in oncology.
Drugs affecting SH3BP5-AS1 would suppress its expression or disrupt its ceRNA activity to restore sensitivity to gemcitabine (mechanism primarily by inhibiting its sponge activity for miR-139-5p and downstream Wnt pathway activation)
