SHANK2 antisense RNA 3 (SHANK2-AS3)

Target
SHANK2-AS3
Molecular classification
Long non-coding RNA (lncRNA), Other (non-protein coding RNA)
01

Overview

SHANK2 antisense RNA 3 (SHANK2-AS3) is a long non-coding RNA transcribed in the antisense orientation to the SHANK2 gene. It is non-protein coding and acts to regulate SHANK2 mRNA stability and translation in neuronal cells. SHANK2-AS3 is upregulated in patients with autism spectrum disorder, leading to reduced SHANK2 protein expression and notable changes in neuronal morphology, including decreased neurite outgrowth, suppressed neuronal proliferation, and increased apoptosis. These functional changes potentially contribute to neurodevelopmental pathologies, particularly autism spectrum disorder. The gene is also known as C11orf76, and is annotated with several aliases in genomic databases. To date, SHANK2-AS3 is not characterized as a druggable therapeutic target, nor is it directly targeted by known pharmacological agents[1][4][6].

02

Mechanism of action

Acts as an antisense long non-coding RNA that binds to SHANK2 mRNA to form double-stranded RNA and suppresses SHANK2 gene expression at the post-transcriptional level

03

Biological functions

Regulation of gene expression (specifically downregulation of SHANK2 protein-coding gene)Modulation of neuronal proliferation and apoptosisMay affect neuronal architecture, especially neurite outgrowth
04

Disease associations

Neurodevelopmental disease (notably implicated in Autism Spectrum Disorder)
05

Safety considerations

None identified in the literature
06

Biomarkers

Upregulation in autistic patients has been observed, thus it may have biomarker potential in the context of Autism Spectrum Disorder
