Overview

SHC adaptor protein 4 (SHC4) is a member of the Shc family of adaptor proteins, which are intracellular molecules lacking intrinsic enzymatic activity and containing phosphotyrosine-binding (PTB) and Src homology 2 (SH2) domains[1][2]. SHC4 functions by mediating protein-protein interactions, transmitting signals from activated cell surface receptors (such as receptor tyrosine kinases) to intracellular signaling cascades, including the Ras/MAPK pathway. It has roles in promoting cell proliferation, differentiation, migration, apoptosis, and oxidative stress responses, and is implicated in tumor development and neurological function[1][2]. SHC4 is considered a potential therapeutic target, but currently there are no known drugs that target it directly or established biomarkers tied to its activity[2]. Dysfunction or deregulation of SHC4 and related proteins has been implicated in cancer, neurodegenerative diseases, and behavioral disorders[1][2].
