SHC SH2 domain-binding protein 1 (SHCBP1) is an intracellular signaling adaptor protein that specifically binds to the SH2 domain of the SHC1 protein. It is primarily localized in the nucleus and cytoplasm of proliferating cells, including cancer cells, stem cells, and lymphocytes, but is largely absent in terminally differentiated tissues. SHCBP1 plays a central role in the regulation of multiple intracellular signaling pathways, including RAS, PI3K/AKT, Wnt/β-catenin, and HER2-PLK1, and is involved in cell cycle progression, inhibition of apoptosis, and promotion of migration and invasion in tumor cells. Overexpression of SHCBP1 is observed in a broad spectrum of cancers and correlates with poor outcomes, making it an important candidate as both a diagnostic/prognostic biomarker and a potential therapeutic target. No currently approved drugs selectively target SHCBP1 in humans; however, research efforts are ongoing to develop inhibitors or RNA-based therapeutics to modulate SHCBP1 activity in cancer therapy.
