The Shigella flexneri 2a O-antigen is a serotype-specific polysaccharide component of the lipopolysaccharide (LPS) on the outer membrane of S. flexneri serotype 2a, a major cause of bacillary dysentery. The O-antigen is critical for bacterial virulence, immune evasion, and serospecificity. It is a branched pentasaccharide composed of rhamnose, N-acetylglucosamine, and glucose, variably modified by glucosylation and O-acetylation. Chain length, regulated by Wzz proteins, affects motility and complement resistance. The O-antigen is a target for protective antibodies and a basis for conjugate vaccine development.
