The O-specific polysaccharide (O-antigen) of *Shigella flexneri* is a key component of the bacterium’s outer membrane lipopolysaccharide (LPS). It plays a critical role in serotype determination, immune recognition, and pathogenicity. The basic repeating unit for most serotypes is a tetrasaccharide composed of three L-rhamnose residues and one N-acetylglucosamine residue. Variations arise from chemical modifications such as glucosylation, O-acetylation, and phosphoethanolamine addition, which are responsible for generating different antigenic determinants that define each *S. flexneri* serotype. It is synthesized via the Wzy-dependent pathway. The O-antigen helps *S. flexneri* evade immune detection and is a target for phage therapy and diagnostic tools.
