Overview

The lipopolysaccharide (LPS) O antigen of *Shigella sonnei* is a surface-exposed polysaccharide that forms the outermost part of the bacterial LPS. It plays a crucial role in pathogenesis, immune evasion, and serotype specificity. The O antigen is composed of repeating disaccharide units of 2-acetamido-2-deoxy-L-altruronic acid (L-AltNAcA) and 2-acetamido-2-deoxy-L-fucose. Unlike other *Shigella*, *S. sonnei* expresses its O-antigen both as part of classical LPS and as a high-molecular-weight group 4 capsule (G4C). The genes encoding biosynthesis/export reside on the pSS virulence plasmid.
