Shigella sp. PIB outer membrane receptors refer to the surface-exposed proteins of the Peristaltic contraction-Inhibiting Bacterium (PIB), a novel strain of the genus Shigella identified as a causative agent of intractable functional constipation (IFC) (Chen et al., 2022, JCI). These receptors, which include major outer membrane proteins such as Outer membrane protein C (OmpC) and Outer membrane protein A (OmpA), are essential for the bacterium's survival, nutrient uptake, and interaction with the host intestinal environment (Qiu et al., 2022, PMC). The primary pathological mechanism of Shigella sp. PIB involves the constant production of docosapentaenoic acid (DPA), which activates host free fatty acid receptor 4 (FFAR4) on nitrergic ganglia in the colon, leading to nitric oxide release and the inhibition of intestinal peristalsis (Wang et al., 2025, PNAS). Consequently, these outer membrane receptors have become significant therapeutic targets for the development of PIB-specific bacteriophages, which bind to the receptors to initiate bacterial lysis and clear the infection. Research into these receptors is critical for developing targeted antimicrobial strategies and diagnostic tools for patients suffering from refractory gastrointestinal disorders linked to this specific pathogen.
