Shisa family member 7 (SHISA7, also known as CKAMP59) is a transmembrane auxiliary protein that associates with AMPA-type glutamate receptors (AMPARs) at the postsynaptic density in neurons[1][3]. It modulates the channel kinetics of AMPARs, impacting their desensitization rates, steady-state current, and recovery from desensitization, and is necessary for normal induction and maintenance of hippocampal synaptic plasticity, particularly long-term potentiation (LTP), as well as for proper memory recall[1]. SHISA7 is most highly expressed in the brain, particularly the hippocampus, cortex, and striatum, and it co-localizes with postsynaptic scaffold proteins such as PSD-95[1][5]. Genetic deletion studies in rodents suggest that SHISA7 is critical for contextual memory but does not substantially affect anxiety or auditory fear learning[1]. There are currently no drugs or known pharmacological modulators that target SHISA7 directly, and its role as a disease biomarker or drug target has not been established. It is classified as an AMPAR auxiliary subunit rather than a conventional receptor or enzyme.
