Overview

Shroom family member 1 (SHROOM1) is a cytoskeletal regulatory protein expressed in a variety of human tissues including the brain, heart, skeletal muscle, colon, small intestine, kidney, placenta, and lung, as well as in melanomas. It is a component of the Shroom protein family, which is distinguished by conserved actin-binding motifs and an ability to interact with Rho kinase (Rock) to spatially assemble contractile actomyosin networks, underlying the regulation of cell shape and tissue architecture during development. SHROOM1 is localized to the cytoplasm and apical plasma membrane of sodium-transporting epithelial cells, where it participates in cell shape modulation. Its hypermethylation has been implicated in the pathology of osteoarthritis, and recent evidence in genome editing highlights SHROOM1 as a suppressor of homology-directed repair, suggesting potential relevance to therapeutic gene editing strategies. No approved drugs or small molecules directly target SHROOM1, and it is not currently considered a classical therapeutic target such as a receptor, enzyme, or transporter.
