Overview

Shroom family member 2 (SHROOM2, also known as APXL and Shrm2) is a cytoskeletal scaffolding protein widely expressed in epithelial and various organ tissues including the nasal cavity, brain, retina, heart, lung, liver, and kidney. It contains a PDZ domain and two Shroom-specific domains (ASD1 and ASD2) that mediate interactions with actin and Rho-associated kinase (ROCK). SHROOM2 controls cell shape by organizing the actomyosin network at the plasma membrane, regulating the distribution of cytoskeletal components, and antagonizing the epithelial-mesenchymal transition implicated in migration and metastasis. Although mapped to a region associated with ocular albinism, SHROOM2 is not functionally involved in this disorder and is recognized for its role in epithelial cell architecture and migration. Unlike receptors or enzymes, SHROOM2 is a structural protein, not directly druggable, and currently has no established biomarkers or therapeutic indications.
