Overview

Shugoshin-like protein 1 (SGO1) is a highly conserved regulatory protein crucial for the maintenance of centromeric cohesion during both mitosis and meiosis[1][3][4]. It acts as a guardian of chromosomal stability, protecting cohesin complexes that physically hold sister chromatids together, thereby ensuring accurate chromosome segregation and preventing chromosomal instability (CIN)[2][3][4]. SGO1 function is tightly regulated by multiple mechanisms, including phosphorylation-dependent recruitment to inner centromeres (via Bub1 kinase and cyclin-dependent kinase 1) and dynamic interactions with associated partners, such as protein phosphatase 2A (PP2A)[1][3]. Disruption of SGO1 leads to severe mitotic errors, chromosomal instability, and embryonic lethality, and is associated with oncogenesis and multisystem disorders such as chronic atrial and intestinal dysrhythmia (CAID)[2][4]. SGO1 is considered a potential therapeutic target in cancer due to its essential role in chromosome segregation, with overexpression or antibody detection reported in certain breast cancers (NY-BR-85 autoantigen)[2]. No approved drugs are known to directly target SGO1, but interfering peptides and downregulation approaches have been explored preclinically[2].
