Overview

Shugoshin-like protein 2 (SGO2) is a conserved chromosomal protein that plays an essential role in meiotic and mitotic chromosome segregation in eukaryotes. It belongs to the Shugoshin/Mei-S322 family, and its primary function is to maintain sister chromatid cohesion at centromeres, protect the cohesin complex during cell division, and recruit the chromosomal passenger complex (CPC) to specific chromatin domains. SGO2 localizes to centromeres and subtelomeric chromatin, regulating the spindle assembly checkpoint, promoting correct kinetochore-microtubule attachments, and contributing to the formation of highly condensed chromatin at subtelomeres in yeasts and mammals. Loss of SGO2 leads to errors in chromosome biorientation or premature chromatid separation, resulting in infertility in mice and increased risk of chromosomal instability and aneuploidy. While SGO2 is crucial for cell division and genomic integrity, it is not currently a therapeutic target nor a biomarker for disease or therapy monitoring.
