Overview

SIAH2 antisense RNA 1 (SIAH2-AS1) is a long noncoding RNA transcribed antisense to the SIAH2 gene. It is highly upregulated in breast cancer tissues compared to normal breast tissue and is implicated in promoting cancer cell proliferation, migration, and invasion. Mechanistically, SIAH2-AS1 activates the Wnt/β-catenin signaling pathway, driving transcription of key cell cycle and EMT-related genes such as cyclin D1 and c-Myc. Knockdown of SIAH2-AS1 reduces the malignant phenotype in vitro by decreasing Wnt signaling and EMT markers, positioning SIAH2-AS1 as a novel oncogenic driver and a potential therapeutic target in breast cancer. Targeted inhibition of SIAH2-AS1 may represent a new approach for cancer therapy, although no drugs currently target it in clinical practice.
