Overview

Siglec-10 is an inhibitory receptor expressed on various immune cells, including B cells, monocytes, and dendritic cells. It recognizes sialylated glycans, particularly α2–3 or α2–6 linked sialic acids. Upon ligand binding, Siglec-10 recruits SHP phosphatases via its ITIM motifs, leading to the inhibition of immune cell activation. The CD24/Siglec-10 pathway is implicated in tumor immune evasion, making it a potential target for cancer immunotherapy. Soluble ligands like CD52 can also engage Siglec-10 to inhibit T-cell activation. Siglec-10 is a type I transmembrane protein with five extracellular immunoglobulin-like domains and two cytoplasmic ITIMs.

Mechanism of action

Ligand engagement recruits SHP phosphatases via ITIMs to dampen immune cell activation and signaling pathways such as NFATc1 and NF-kB in B cells through SHP1/Grb2 recruitment.
