Siglec-15 is a type I transmembrane glycoprotein that belongs to the Siglec family of sialic acid-binding immune regulatory molecules. It is primarily expressed on osteoclasts, macrophages, and dendritic cells, and its expression can be upregulated in various cancers. Siglec-15 functions as an immune suppressor, inhibiting T cell responses and promoting immune evasion in the tumor microenvironment. It also plays a critical role in osteoclast differentiation and function, contributing to bone resorption. Due to its involvement in both immune suppression and bone resorption, Siglec-15 has emerged as a promising target for next-generation cancer immunotherapies and potential treatments for metabolic bone diseases.
Monoclonal antibodies targeting Siglec-15 are being developed to block its interaction with its ligands and downstream signaling pathways, thereby reversing its immune-suppressive effects and promoting anti-tumor immunity. In bone disorders, targeting Siglec-15 aims to inhibit osteoclast activity.
