Siglec-8 is a member of the CD33-related Siglec family, selectively expressed on eosinophils and mast cells. It functions as an inhibitory receptor, and its engagement induces apoptosis in eosinophils through a signaling cascade involving SFKs, Syk, SHIP1, PAK1, MEK1/ERK1/2, PLC/PKC, acid sphingomyelinase/ceramidase, and Btk. Siglec-8 is a therapeutic target for diseases involving eosinophil accumulation/activation, such as asthma and hypereosinophilic syndromes.
Monoclonal antibodies targeting Siglec-8 induce eosinophil depletion/inhibition via apoptosis.
