SIGLEC8 is an immune-inhibitory receptor selectively expressed on eosinophils, mast cells, and basophils. As a member of the CD33-related Siglec family, it binds to α2,3-linked sialic acids, particularly those containing sulfated galactose residues such as 6'-sulfo sialyl Lewis^x. Engagement of SIGLEC8 induces eosinophil apoptosis and inhibits mast cell degranulation, making it a promising therapeutic target for eosinophilic and allergic diseases.
Agonistic monoclonal antibodies or glycan ligands bind to SIGLEC8, inducing eosinophil apoptosis via Src/Syk/PI3K/PLC/PKC/Rac1-MEK1–ERK/CD11b-CD18 integrin pathways or inhibiting mast cell degranulation.
