Siglec-6 is a member of the CD33-related Siglec family, acting as an inhibitory receptor expressed in placental trophoblasts, mast cells, and memory B-cells. It binds sialic acid-containing glycans, particularly α2–6-linked sialosides. Upon ligand binding, ITIM/ITSM motifs in its cytoplasmic domain recruit SHP phosphatases, dampening activation signals. It also regulates cell migration and adhesion. Siglec-6 is overexpressed in CLL cells and colon cancer, making it a target for antibody-based therapies.
