Overview

Sialomucin core protein 24 (CD164) is a type I transmembrane sialomucin glycoprotein and cell adhesion molecule predominantly expressed on hematopoietic progenitor cells and bone marrow stromal cells[1][3][4][6]. It plays critical roles in regulating hematopoietic stem and progenitor cell proliferation, adhesion, and migration, partly through its interaction with the chemokine receptor CXCR4 and modulation of the CXCL12/CXCR4 axis[1][3]. CD164 is also involved in muscle development by affecting myoblast motility and fusion, participates in early activation events in basophils, and acts as an entry receptor for lymphocytic choriomeningitis virus (LCMV)[1]. Aberrant expression or mutation of CD164 has pathological implications, including stimulating tumor progression and metastasis in several cancers and association with hereditary hearing impairment (DFNA66)[1][3]. CD164 is an extensively glycosylated molecule and serves as a marker for early hematopoietic stem/progenitor cells and basophil activation, and its diverse physiological and disease roles make it a candidate diagnostic and therapeutic target[1].
