Sideroflexin-5 is a member of the Sideroflexin (SFXN) family, which consists of highly conserved multi-spanning transmembrane proteins mainly located in the mitochondrial inner membrane. SFXN5 is primarily characterized as a mitochondrial amino-acid transporter and mediator of citrate transmembrane transport. It does not function as a serine transporter, unlike some other family members (e.g., SFXN1). In brown adipose tissue, SFXN5 may regulate thermogenesis by supporting mitochondrial glycerol-3-phosphate utilization. Current research links the SFXN family to essential metabolic processes such as iron uptake, redox balance, and amino acid metabolism, but SFXN5 remains the least well-studied member. Disease associations include mitochondrial deficiency syndromes and possible roles in neurodegenerative and cancer processes, based on emerging expression data and known implications from other family members. There are currently no approved drugs targeting SFXN5 directly, nor is it an established biomarker or well-defined safety target.
