Overview

The sigma-1 receptor (S1R), also known as sigma non-opioid intracellular receptor 1, is a unique ligand-operated chaperone protein primarily located at the endoplasmic reticulum (ER) membrane, especially in mitochondrial-associated membranes (MAMs). It modulates calcium signaling, lipid dynamics, and the cellular stress response by interacting with proteins such as GRP78/BiP and ion channels, GPCRs, kinases, and NMDA receptors. It has been implicated in neurological disorders, psychiatric conditions, drug dependence, cardiovascular function, and cancer.
