Signal-induced proliferation-associated 1-like protein 1 (SIPA1L1) is a large cytoplasmic protein that acts as a GTPase-activating protein (GAP), particularly for RAP2A, and modulates synaptic function by regulating actin cytoskeleton organization and dendritic spine morphogenesis. It interacts with key synaptic proteins such as DLG4 and components of the ephrin receptor signaling pathway, and is implicated in neuronal morphology and synaptic plasticity. Variants in SIPA1L1 have been associated with certain cancers and autism spectrum disorder. Therapeutic targeting of SIPA1L1 has not yet been established[1][5][6][7].
