Overview

Signal peptidase complex subunit 1 (SPCS1) is an integral, non-catalytic component of the signal peptidase complex (SPC) in the endoplasmic reticulum membrane[1][3]. The SPC is responsible for the cleavage of N-terminal signal peptides from nascent secretory and membrane proteins as they translocate into the ER, permitting their maturation[1][3]. SPCS1 is not essential for enzymatic activity but is involved in processing viral proteins and promoting virion assembly, specifically for flaviviruses such as hepatitis C virus[1][2][4]. SPCS1 plays a role in forming complexes between viral and host proteins required for effective viral particle assembly, though it does not itself directly catalyze peptide bond cleavage[2][4]. Recently, the signal peptidase complex has also been implicated in cellular quality control by supporting the degradation of faulty membrane proteins[6]. SPCS1’s deficiency impairs propagation of certain viruses but is not widely involved in the general regulation of the viral lifecycles of all flaviviruses[4]. No therapeutics directly target SPCS1, and there are no known biomarkers or specific safety concerns attributed to SPCS1 modulation. There is no evidence that SPCS1 is currently considered a primary therapeutic target in human disease.
